Today’s Feature Article at work:

 

"Mental retardation affects a larger number of males in the population than females, and this has been attributed, in part, to the increased proportion of mutations in genes on the X chromosome. The number of causative mutations leading to X-linked mental retardation (XLMR) is unknown, and will remain so until all of the genes on the X chromosome have been fully investigated. While this research is ongoing, screening for the XLMR remains a challenge. Here we describe the direct detection of non syndromic X-linked Mental Retardation (MRX) variants using the Applied Biosystems 3500xL Genetic Analyzer".

The report could have simply stated, "because they have penises."

 

Do I remember correctly/accurately that all human embryos begin essentially female, but males are derived by hormone introduction?

LOL! Very similar, I remember dropping my military ID just as I was entering the post (walking) after dropping some window pane and taking about 5 min to get it picked up.....quite disconcerting then, funny now.

hey mito, how much big dye you using? I'm trying to reduce our consumption

 

Using a BigDye Terminator v3.1, we were able to get a decent signal with as low as 1/10 of what is suggested by ABI, i.e.

0.4 ul in the total reaction volume of 10 ul. With a little bit of tweaking, I would say - 1/20, BUT the ratio of your primer and DNA template should be just right, the PCR product uber clean, etc. You do the math: your time versus $$$. We have also switched to formamide as an injection carrier.

Do you use the bdt buffer or just toss it (or make your own)

 

We used to make our own 10x BDT:

400 mM Tris-HCl, pH = 9.0

10 mM MgCl2

 

Now, we are using the DNA Sequencing Dilution Buffer III from TekNova (cat. # D1301), super cheap - $40.00 for 500 ml.
